Objective: Alongside its role as a potent vasoconstrictor in the cardiovascular system, endothelin-1 (ET-1) is involved numerous physiological processes. Its expression thought to be an important function for both breastfeeding and newborns since it occurs especially during pregnancy lactation. In this study, we projected effects of ET-1 on mammary gland physiology employing transgenic mice. Materials Methods: compared glands mice with control different states (adolescence, pregnant, lactation involution). Parallel sections were prepared from microscopic examinations performed hematoxylin eosin (HE)-staining. Western blot techniques used analysis signal proteins that have functions development metabolism (STAT5, AKT, STAT3). Genes known play key metabolic activities (WAP, beta casein, GLUT-1, SRBF-1 Claudin 8, IL-6, LIF) analyzed real-time PCR techniques. Results: It was determined developed pathological conditions such secretory damage, lactational failure early involution, respectively. Our western studies showed STAT5 activation not affected by process transgenic group, but decrease AKT protein level detected. We detected STAT3 seen involution only 3rd day addition, found WAP suppressed due high LIF increased significantly, analysis.
